Single-Triggered Trimeric Prodrugs

This article, published in 2004, describes the first prodrug system with the ability to release three different anticancer drugs in the presence of antibody 38C2.  The authors covalently bind Campthocein, Doxorubicin, and Etoposide to a single enzyme substrate and show the release of all drugs after addition of antibody 38C2.  Antibody 38C2 cleaves the enzyme substrate and triggers retro-aldol and retro-Michael reactions that in turn cleaves the drugs.
